Objective

  • Make measurement samples from the new stocks of malachite green prepared
  • Measure UV-Vis and fluorescence

Data

  1. Stock solutions made by the group G: 0.3ppm, 1ppm, 3ppm, and 6ppm
    1. 0.30885 ppm (0.52711 g of 29.3 ppm Malachite green stock, water added to a total mass of 50.0 g)
    2. 1.00500 ppm (1.71704 g of 29.3 ppm Malachite green stock, water added to a total mass of 50.049 g)
    3. 3 ppm (5.1194 g of 29.3 ppm Malachite green stock, water added to a total mass of 50 g)
    4. 6 ppm (1.023 g of 29.3 ppm Malachite green stock, water added to a total mass of 10 g)

Notes

  • We measured UV-Vis, followed by fluorescence
  • our concentrations(0.3ppm and 1ppm) were too dilute and now signal was picked up by fluorescence
  • We then made two more solutions(3ppm and 6ppm) of higher concentrations and took fluorescence of them, but again, no signal was picked up
  • For the 6ppm we ran out of 50 mL falcon tubes and used 15 mL ones instead and calculations were adjusted to make 10 g of the solution instead of 50.
  • Dr. Hartings then prepared Rhodamine samples that were known to fluoresce
    • Parameters Used:
      • Start(nm): 565 End(nm): 700 Excitation(nm): 555
      • Ex slit(nm):5 Em slit(nm): 50 Scan Speed(nm/min): 200
