Objective

  • Synthesis of Bovine Serum Albumen Conjugated Gold Nanoparticles using H2O and Acetate (pH=4.3) as seperate buffers. This is a repetition of the experiment performed on August 31st, 2011.

Description

  1. Prepared 10 mL aqueous mixtures of 2.5mM HAuCl4 and 15 μM Bovine Serum Albumin(BSA) in H2O.
  2. Prepared 10 mL aqueous mixtures of 2.5mM HAuCl4 and 15 μM Bovine Serum Albumin(BSA) in 50mM of Acetate (pH=4.3) as the buffer.
  3. Placed each reaction mixture in an 80°C water bath.
  4. Obtained UV-Vis Spectra in the wavelength range of 200-800nm at 30 minute intervals.

Data

  • 2.5 mM of HAuCl4 and 15 μM of BSA were the two stock solutions used.
  • To prepare BSA (1.5μM)and HAuCl4 (0.25mM), the equation M1V1=M2V2 was used in the following calculation:
  • Calculation

2.5mM*V1 = 0.25mM*10ml

V1= 1ml of HAuCl4


0.015mM*V1 = 0.0015 mM*10ml

V1= 1ml of BSA

  • 1ml of HAuCl and 1ml of BSA were placed in each of the two test tubes.
  • The graph below is the UV-Vis Spectra of HAuCl4/BSA/H2O obtained in the wavelength range of 200-800nm.

  • The graph below is the same as the graph above it, but is zoomed in at 550nm.

  • The graph below exhibits the change in absorbance at 550nm over a period of 2 hours.

  • The figure below is the UV-Vis Spectra of HAuCl4/BSA/Acetate obtained in the wavelength range of 200-800nm.


  • The graph below is the same as the graph above it, but is zoomed in at 550nm.

  • The graph below exhibits the change in absorbance at 550nm over a period of 2 hours.

Notes

  • Both mixtures contained purple fibers floating in a clear solution.
  • The spectra showing the formation of conjugate gold nanoparticle formation over time at 550 nm in either H2O or acetate buffer indicated conjugate gold nanoparticle synthesis because of their increasing trendlines.
  • The mixture in acetate, had a greater slope than the water mixture indicating that the rate of conjugate gold nanoparticle synthesis was greater in the acetate mixture.
