Objectives

  • If the concentrated original nanofiber method worked with the scaffolds,
    • Take pictures against white background
    • Print 7 of the scaffold setup that worked and make more nanofibers according to yesterday's procedure
    • Start tests
  • "Brute Force" method
    • For each nanofiber solution, there is one third of the tall scaffold
    • Place one tube worth of nanofibers on one of the thirds (repeat for other eight scaffold thirds)
    • Allow nanofibers to dry on the scaffolds overnight
  • Determine method of quantification for nanofibers on the scaffolds

Nanofiber Results

100:1 0.5mM Au:Myoglobin nanofibers alone<br.> <br.> 100:1 0.5mM Au:Myoglobin nanofibers on 1 tall scaffold<br.> <br.> 100:1 0.5mM Au:Myoglobin nanofibers on 4 tall scaffolds<br.> <br.> 100:1 0.5mM Au:Myoglobin nanofibers on 1 extra tall scaffold<br.> <br.>

Nanofiber Retention Tests

  1. Put coated scaffold in PBS buffer
  2. Put coated scaffold in ethanol
  3. Put coated scaffold in water at 35C for 30min
  4. Put coated scaffold in water and sonicate for 30min
  5. Put coated scaffold in 0.1M HCl solution
  6. Put coated scaffold in water
  • Pictures will be taken of sonicated and 35C after sonication and heating are complete
  • Pictures will be taken of all solutions approximately every 2 days (Friday, Monday, and Wednesday)

Nanofiber Retention Test Results: Sonicated and Heated

  • Both samples appear unchanged
