For plate 1, what does "some background DNA" mean? Did you mean to say some background colonies?

Plate Number Contains Comments
1 vector only some background DNA, this was expected
2 insert only nothing, good result
3 both vector and insert (1:1 V:I) nothing, bad result
4 both vector and insert (2:1 V:I) 1 colony, funny result but still bad
5 nothing but E coli cells nothing, good display of sterile technique

Growth

We took that one colony and used it. We figured we might as well. I didn't want to waste all that effort I had put in.
Method:

  1. I plucked the colony
  2. Placed colony into LB media
  3. Place tube into shaker overnight
  4. Watch it grow.
