The spec should be ready to read all the wells in a 96 well plate.  If the Green Read button on the top toolbar is missing, it means the instrument is Offline. To get it back Online, Click the Spectromax 190 icon on the left of the top toolbar. An Instrument Connect popup window should appear with an hourglass.
