• Follow the Thermo Fisher Phusion Hot Start II DNA Polymerase instructions. DNA can be from a miniprep of from a colony. If assaying a colony, one can: i) pick a colony, resuspend it into 20 ul of water/LB and pipette 1 ul into the PCR mix (recommended) or ii) simply "infect" the PCR mix with the picked colony by pipetting up and down.
