  • E coli culturing
    • Biosafety Hood UV-irradiated for 20 mins, turned off and left for 5 mins (SOP)
    • 3/12 E coli culture removed from shaking incubator (incubate 16 hrs)
    • Under the hood, one loopful of E coli streaked (quadrant streaking method) onto LB(A) plate (as immediate culture)
    • Under the hood, one loopful of E coli streaked (quadrant streaking method) onto LB(A) slant (as stock culture)
    • 2X E coli plate and slant incubated at 37 C overnight (time start: 1020 hrs)
  • Medium preparation
    • Repeat of 3/12 procedure [1]

Notes

  • Always Parafilm petri dishes when storing
  • Overturn dishes upon storing (for incubation, overturn only when no moisture is visibly found)
