Overall the PCR Reaction Report was simple. Before the lab we had an online prelab task to complete which basically showed us how to pipette online. When it came down to the lab we had the knowledge already to complete the pipetting experience because of the prelab. Before my group began pipetting we first labeled our tubes which was a little difficult because the sharpie wasn't showing to well at first. After we got done with pipetting we were a little worried because we noticed there was left over liquid in the bottom of the tubes, we then agreed to go over and pipette the rest of the liquid in the right tubes.
Fluorimeter Procedure
Imaging set-up
First we placed a smartphone the cradle at a right angle form the slide. We then adjusted the height of the fluorimeter by using plastic trays so that the smartphone takes pictures of the droplets at the desired angle, this also helps file out the image. We then adjusted the distance between the smartphone/cradle and the first two rows of slides so that the image can be close as possible without it being blurry.
Placing Samples onto the Fluorimeter
Place the fluorimeter on the table and turn it on.
Then place the slide in the fluorimeter with the smooth side facing down
Next we set the camera up by setting the timer to three seconds
Then place 80 uL of syber green solution on the slide using a micropipette
Then we placed 80 uL sample/calibration solution and injected it on top
Next adjust the slide so the light is directly on the drop
Place the light box over the fluorimeter along with the smartphone while keeping one flap up
Then close the flap before the picture is taken
Once the picture was taken remove 160 uL from the slide and properly disposed of it
Once again adjust the slide so that the light goes through the circles on there
Repeat all the following steps above to collect all five measurements
