Objective Begin growth of BL21(DE3) E.coli with mutated GFP plasmid (D1C) for expression. Description LB was kindly prepared by Dr. Hartings for this expression. 25μL of 100mg/mL ampicillin was added to 25mL of LB (four of these). One colony was added to each of these flasks. The flasks were incubated overnight at 37°C at 250rpm. Notes There was no growth overnight. The plate that the colonies came from was probably too old, and also may have been contaminated.
