Dye Addition

  • Preparation of Rhodamine 6G stock solution
Rhodamine 6G '
Molecular Weight (g/mol) 479.02
mass(g) 2.3951
volume stock (L) 0.01
Concentration (M) 0.5
Real concentration of Rhodamine used '
concentration (M) 0.005
volume stock (L) 0.1

Dye addition to samples was done at a range of concentrations from 20 μM - 200 μM and was added to 1mL of sample. The samples need at least 24 hours in order to soak in the dye.

  • Fluorescence of the water sample which the nanoparticles/ rhodamine will be analyzed to determine the total R6G concentration adsorbed by the hydrogel complexes.
Sample Number Internal Conc. Dye in each sample (M) volume of dye added in uL
1 0.00002 0.000004 4
2 0.00004 0.000008 8
3 0.00006 0.000012 12
4 0.00008 0.000016 16
5 0.0001 0.00002 20
6 0.00015 0.00003 30
7 0.0002 0.00004 40

Samples were placed in eppendorf tubes and manually shaken/ stirred for approximately 2 minutes each in order to mix dye and prepare samples for absorption/ crosslinking. Samples were left overnight.

NOTES

    • From UV-Vis spectra collections, it was determined that the true concentration of the R6G that was used was: 398.7 uM
