Objective

  • Prepare 0.1M buffer solutions of pH 10, 11, & 12
  • Make a 10mM HAuCl4 solution and a 100mM AA solution (using an AA with a basic R group)
  • Prepare Buffer for Bradford Analysis
  • Prepare Buffer for Dialysis
  • Prepare Buffer for SDS-PAGE

Description

  • Prepare 0.1M bugger solutions of pH 10, 11, & 12
Following the previous AA mass calculations. Buffers at pH 10, 11, & 12 were used instead of water.



  • Make a 10mM HAuCl4 solution and a 100mM AA Solution (using AA with a basic R group)
10mM calculation HAuCl4 (339.785g/mol) solution
(10E-3M)*(0.25L)=0.0025moles
(0.0025moles)(339.785g/mol)=0.8495 grams
100mM AA solution (Arginine = 174.2g/mol)
(100E-3M)*(0.04LL)=0.004moles
(0.004moles)(174.2g/mol)=0.70grams



  • Prepare Buffer for Bradford analysis
1L of 50mM Tris, 50mM NaCl pH7.5
Tris (121.14g/mol) mass calculations
(1L)*(50E-3M)=0.05mol
(0.05mol)*(121.14g/mol)=6.057grams
NaCl (58.44g/mol) mass calculations
(1L)*(50E-3)=0.05mol
(0.05mol)(58.44g/mol)=2.922grams
pH measurements in order
pH=10.34; 9.55; 9.08; 8.53; 8.11; 7.77; 7.47



  • Prepare buffer for Dialysis
