Objective

To test the procedures from 12/06/11 with different ratios of Au/BSA. Test ratios of 100 and 35.

Description

  • Create solutions for ratio of 100 Au:BSA
    1. Using 17.7 μM BSA stock solution and 2.5 mM HAuCl4 stock solution, combine 0.5 mL gold solution and 0.0706 mL BSA solution.
    2. Dilute with 5 mL of water.
    3. Repeat to make a second solution.
    4. Cover with tin foil.
  • Create solutions for ratio of 35 Au: BSA
    1. Using the same stock solutions, combine 0.5 mL gold solution and 2.02 mL BSA solution.
    2. Dilute with 5 mL of water.
    3. Repeat to make a second solution.
    4. Cover with tin foil.
  • Put all solutions into the oven at about 75°C.
  • Leave one of each ratio concentration in for the duration of lab (about 3 hours).
  • Take one of each ratio concentration out for 10 minutes every half hour for 3 hours.

Data

Solution 1: Au/BSA = 35, left in the oven

Solution 2: Au/BSA = 35, taken out of the oven every 30 minutes

Solution 3: Au/BSA = 100, left in the oven

Solution 4: Au/BSA = 100, taken out of the oven every 30 minutes
