Objective

All of the reaction mixtures from Wednesday's lab produced a homogeneous solution of nanoparticles (AuNPs) instead of fibers (AuNP fibers).

The most likely reason is that the balance did not properly record the mass of the lysozyme. Today I am going to remake the lysozyme solution and synthesize several 1 mL samples of AuNP fibers.

Description

Lysozyme stock solution

  1. Mass of lysozyme = 15.21 mg
  2. Molar mass of lysozyme = 14307 g/mol
  3. volume used = 50.0 mL
  4. Lysozyme concentration = 21.3 μM

For these samples, I am using the gold stock solution from Wednesday and HPLC-grade water.

Trial solutions to determine if the stock is good

  1. Au:Lysozyme ratio = 45
    1. Volume of Au stock = 422 μL
    2. Volume of Lysozyme stock = 1,297 μL
    3. Volume of Water = 3,281 μL
    4. Place in a test tube and cap with foil
  2. Au:Lysozyme ratio = 50
    1. Volume of Au stock = 422 μL
    2. Volume of Lysozyme stock = 1,167 μL
    3. Volume of Water = 3,411 μL
    4. Place in a test tube and cap with foil
  3. Au:Lysozyme ratio = 55
    1. Volume of Au stock = 422 μL
    2. Volume of Lysozyme stock = 1,061 μL
    3. Volume of Water = 3,517 μL
    4. Place in a test tube and cap with foil
  4. Au:Lysozyme ratio = 60
    1. Volume of Au stock = 422 μL
    2. Volume of Lysozyme stock = 972 μL
    3. Volume of Water = 3,605 μL
    4. Place in a test tube and cap with foil
  5. Au:Lysozyme ratio = 65
    1. Volume of Au stock = 422 μL
    2. Volume of Lysozyme stock = 898 μL
    3. Volume of Water = 3,680 μL
    4. Place in a test tube and cap with foil
  6. Au:Lysozyme ratio = 70
    1. Volume of Au stock = 422 μL
    2. Volume of Lysozyme stock = 834 μL
    3. Volume of Water = 3,744 μL
    4. Place in a test tube and cap with foil

Samples for Students (1 mL in eppendorf tubes)

  1. Au:Lysozyme ratio = 45
    1. Volume of Au stock = 84 μL
    2. Volume of Lysozyme stock = 259 μL
    3. Volume of Water = 656 μL
    4. Place in a test tube and cap with foil

Data

All of the samples (ratios: 45, 50, 55, 60, 65, 70) produced fibers with the new protein stock. I am going to go ahead and make the student samples with this stock.

Checked on Monday, September 8th 2015 Most of the samples produced AuNP fibers. Some produce purple solutions with some fibers crashing out of solution. I think that this is due to effects from the plastic. Will have to look in to this. But, going forward, we will only use the samples that clearly have only fibers in them.
