Bradford Analysis

  • A set of standards with known concentration were created from BSA stock solution (dissolved in saline) in order to construct a standard curve according to their Bradford values which could be useful to determine the concentration of a protein that we were assaying. The 6 standard solution with a concentration of 1 µg/mL – 20 µg/mL BSA and a total volume of 1 mL were prepared containing 200 µL Bio-Rad Protein Assay reagent and a suitable amount of Tris/NaCl buffer. The UV-visible spectra of blank solution (without protein) and standard solution were obtained within 1 hour of preparation at 400-800 nm. Based on the calibration curve created from obtained absorbance, the concentration of original BSA and amino acid stock solution were determined.
