Procedures

  • Dissolve ZnPPIX made 10/09/13 in DMF
  • Filter ZnPPIX solution to remove any more zinc or porphyrin impurities
  • Allow to dry overnight

Data

  • 0.0146g ZnPPIX dissolved in 13mL DMF
  • MW ZnPPIX: 626.032g/mol

(0.0146g)×(626.032g/mol)×(1/0.013L)≈1.793mM<br.>

  • ε412 ZnPPIX in phosphate buffer: 87.4mM/cm
    • (from "Metal and redox selectivity of protoporphyrin binding to the heme chaperone CcmE")

Notes

Next week:

  • Re-dissolve ZnPPIX in DMF
  • Prepared several dilutions (100x, 1000x, 10000x?)
  • Take UV-vis measurements
  • Use Beer's law to determine concentration
