Objective Confirm that PCR worked and product is consistent with expectations. Also confirm that miniprepped plasmid is consistent with expectations. Bench work Analytical digest of pCMV-SPORT6+BSA] with XmnI Daniel Catt did this 8.9μL 158μg/mL pCMV-SPORT6+BSA + 1μL 10X NEB4 buffer + 0.1μL 100X BSA + 1μL 20kU/mL XmnI →1h @ 37°C →4°C Analytical minigel 1.2% Agarose gel (~20 mL with small-tooth comb) Mix samples with 6X DNA loading buffer and load: 10μL 1KB ladder 10μL XmnI-digested plasmid from step 1 5μL PCR product from yesterday, step 3 Results and Conclusions Results are consistent with expectations.
