Objective

  1. Re-redo our experiment to test the uptake of gold nanoparticles
  2. Create lysozyme nanoparticles

Protocol

AuNP Uptake Setup
The experiment on 02/10/2016 failed because we neglected to cover the jars with foil or Parafilm, so today we are going to do that.

Today, 20mL of AuNP stock at a 25% dilution was used of both the gold and red AuNPs in small jars. Again, plants with extensive root systems were harvested and added to the small jars containing the AuN solutions, and the jars were covered with foil. They were left out on the lab bench to be checked in 24hours.


Lysozyme Nanoparticle Setup
100mL stock solution was made using the following steps:

  1. 7.72mg of lysozyme was added to 25mL of water in a volumetric flask (2.16M). The solution was then transferred to a 250mL beaker.
  2. 0.84mL of 1% by weight HAuCl4 stock solution was added (made on 01/27/2016)
  3. 74.16mL of water was added, bringing the final solution volume up to 100mL.

The solution was then covered with foil and placed in the oven for 4 hours at 80 degrees celsius.

Results

We made fibers! Not nanoparticles. But we saved the fibers to instead incorporate into a film for future experiments.

Also, our java ferns finally showed promising results. The purple nanoparticles showed slightly better uptake so from now on we are synthesizing and using just the gold nanoparticles. Results are shown below:
