Ligation and Tethering We are doing the ligation reaction and tethering today. We started off with the ligation, where we mixed together 25μL of the pBR322, 4.25μL anchor, 2μL of the adapter, 5μL of the buffer, 1μL of DNA ligase and 6.25μL of the DI water.
