I'm doing with and without perfect match to see if this affects anything.
Results
The reaction failed so I will try again without perfect match.
Anthony Salvagno 14:55, 12 April 2011 (EDT):The reaction failed again and so I lowered the temperature on the thermal cycler for the annealing phase to see if that fixes the problem.
