When cutting with EarI, there are two restriction sites. One is at bp#2351 and the other is at bp#4155. The plasmid is 4361bp long. This gives us lengths of 2557 and 1804bp. When ligating this to the unzipping adapter only the big piece has the overhang needed for the ligation to work.
