So yesterday I ligated the anchor to the hairpin adapter, and then digested the hairpin with NotI. Today I PCR purified (QIAQuick) the digested sample because the hairpin fragment would just go through the membrane of the column. Then I ran a ligation of this construct to the Notsamples.

Next time I will gel purify these guys.
