Just as we did with the NotI digestion we did a test digestion with SapI. We took a small sample of all the tubes and digested. I then did a gel to see if there were any SapI sites within our gDNA fragments. The results are inconclusive because some of the fragments didn't digest. But it looks like we are going to do a good digestion of many of the same tubes as the NotI digest. They are:

  • D
  • 14
  • 16
  • 21

Tubes D, 16, and 21 are the same as NotI digest.

I also ran out of DNA so we are gonna save the last little bit to do a transformation.

I also sent out 10uL of tubes D, 2, 14, 16, 21 for sequencing
