You will determine whether the N15 protelomerase parts are functional or not. In your experiments, you'll cotransform protelomerase-expressing plasmid and a plasmid containing the tos site. Assuming you get stable replication, you'll miniprep and map the tos plasmid to determine whether it was linearized or not.
If the plasmids don't stably cotransform, you will perform enrichment experiments by mixing tos plasmid with an RFP-expressing plasmid without tos and determining whether cells containing N15 protelomerase enrich for plasmids lacking tos.
