Jasonk 07:23, 14 September 2006 (EDT): Why do the 2 part plasmids need to have different resistances from each other? Couldn't you have 2 parts on Amp plasmids and transform into a Kan construction vector?

  • Reshma 16:24, 3 January 2008 (CST): Yes you can. The example just demonstrates that even if the 2 part plasmids have different resistances, you can still use a destination plasmid with a third resistance. Thus, you only need 3 vectors with 3 different resistance markers.
