colony pcr pick half of an isolated colony with a toothpick of pipet tip and inoculate 25 µl TE; store the plate parafilmed at 4ºC. boil for 5 min. spin down at 13k for 1 min. use 2 µl as template for subsequent pcr. run out on a gel with respective control. pick other half of the positive clones and inoculate liquid o/n culture for miniprep.
