Tris-Buffer

Materials

  • MilliQ Water
  • TrisBase
  • HCl


Procedure

Tris-Buffer can prepared at various desired pH values. This procedure covers how to prepare a Tris-Buffer of pH 7.0, 7.5, and 8.0 respectively. Each protocol prepares 2L of solution. In addition, each Tris-Buffer requires the addition of 242.2 g of TrisBase.

First, add 242.2 g of TrisBase to 2L of MilliQ Water. Then in order to prepare desired pH add 37% HCL accordingly:

  • pH 7.0, 150-155mL HCl
  • pH 7.5, 120-125mL HCl
  • pH 8.0, 80-85mL HCl
