31.08.2009 1. We made digestions. The following amounts are added to microtubes; 2 , Buffer 3 µl Xba1 3 µl DNA 18 µl water 6 µl =30 µl BB, Buffer 3 µl EcoR1+Pst1 1.5+1.5 µl DNA 20 µl water 4 µl =30 µl 2. Electrophoresis order; 1)leader / BB control / BB / 2 control / 2 3. Gell extraction for 2 and BB were made.
