To test and see if the plasmids in these colonies are our intended colonies (ligated insert + backbone), we performed colony PCR. After PCR, we plan to run a gel. This will allow us to determine the lengths of the DNA fragments that the primers cut at. Since we know the size of our insert, we will be able to tell if the DNA fragment is our insert or not.
