02/13/2014 Golden gate colonies 1-4 did not grow Realized the Lux boxes were not correct. Ordered new ones 5-6 grew >10 small colonies Picked two from each plate into 3mL LB+amp Positive control grew hundreds of medium sized colonies Got some more K12s from Kylie to make competent cells. Picked more DH5αTs to make more competent cells
