Zinc Fixation Protocol


  • 1000 mL 0.1M Tris buffer, pH 7.4
  • 0.5 g Calcium Actate
  • 5.0 g Zinc Acetate
  • 5.0 g Zinc Chloride

Mix to dissolve, do not adjust pH (it should be about 6.5-7.0).

I) Place freshly dissected tissue (3 mm or thinner) in Zinc-Fixative and allow to sit at room temperature 6-48 hours.

II) Dehydrate tissues as follows, all at room temperature:

  • one hour 70% ethanol
  • one hour 95% ethanol
  • one hour 100% ethanol
  • one hour 100% ethanol

III) Clear tissues as follows, both at room temperature:

  • 50 minutes in Xylene
  • 50 minutes in Xylene

IV) Follow with paraffin embedding.
