03/08/10: Bacteria growth Started growing at 10:00. Had 2 x 250 mL M9 media (one for 17°C and one for 37°C) and 1 50 mL 2xYT+NPS as control. Inserted 1:100 volume of starter solution, which seemed good. Started checking OD: The table's caption Time 17°C 37°C ctrl/37°C 10:00 0.092 0.096 0.094 16:00 0.100 0.09 1.14 18:40 0.100 0.08 - 21:10 0.098 0.096 - 10:10 (4/8/10) 0.100 0.1 - At 16:00 I took the ctrl flask from 37°C, added 6 μL 840 mM IPTG and inserted to 17°C to induce expression O/N. M9 flasks did not show any growth.
