Try re-dissolving failed primers AV10126 (B03 B04) and AV10094 (C05 C06) that failed previously Reagents 1x conc. 6 h2o 6.75 - 40.5 10x 1 10x 6 dye 1 10x 6 dNTP 0.2 10 mM 1.2 Forward 0 10 uM Taq 0.05 5 U/um 0.3 DNA 1 5 ng/ul TOTAL 10 Cocktail per vial 9 3m 95C (10s 95C, 60C 1m)x35 first three wells -- a test extraction (1ul of single rotifer in 40 ul GeneMatrix) second three wells, re-duluted primers, with a positive control in the first well. need to re-design primers
