Media

  • In a 500mL Erlenmeyer flask add 200mL of dH2O and 4 boiled wheat grains.
  • Add 200μL of CH or 200μL of PJ solution
  • Let stand at room temperature 24 hours. Use a small piece of tin foil to cover the top of the flask (not tightly, you want air to flow).
  • The next day inoculate with A. proteus and maintain at room temperature in the dark.
  • Optional: You can also try to use the P. caudatum media but use 4 wheat grains.

Subculture

When you have a healthy culture going (about 1-2 weeks after inoculation) you can subculture once a month (approximately) in fresh media with new wheat grains. Remember to prepare the media the day beforehand.

Notes

  • Cells collect on the bottom around the wheat grains. Very sensitive to higher temperatures.
