Media In a 500mL Erlenmeyer flask add 200mL of dH2O and 4 boiled wheat grains. Add 200μL of CH or 200μL of PJ solution Let stand at room temperature 24 hours. Use a small piece of tin foil to cover the top of the flask (not tightly, you want air to flow). The next day inoculate with A. proteus and maintain at room temperature in the dark. Optional: You can also try to use the P. caudatum media but use 4 wheat grains. Subculture When you have a healthy culture going (about 1-2 weeks after inoculation) you can subculture once a month (approximately) in fresh media with new wheat grains. Remember to prepare the media the day beforehand. Notes Cells collect on the bottom around the wheat grains. Very sensitive to higher temperatures.
