Loading dyes and DNA markers are stored in a gel room refrigerator.

6x Xylene Cyanol Loading Buffer for Electrophoresis


This buffer will have only the light blue than runs slower on the gel. This protocol will make 40ml of 6x solution. Adjust to your needs if necessary.

  1. 100mg Xylene-cyanol
  2. 6g of Ficoll (Type 400)
  3. Add water to 40 ml

Dissolve well. It takes a while. An option is to shake at 37 degrees overnight.

6x Bromophenol Blue Loading Buffer for Electrophoresis


This buffer will have only the dark blue than runs faster on the gel. This protocol will make 40ml of 6x solution. Adjust to your needs if necessary.

  1. 100mg Bromophenol Blue
  2. 6g of Ficoll (Type 400)
  3. Add water to 40 ml

Dissolve well. It takes a while. An option is to shake at 37 degrees overnight.

Another protocol (Liron Shalom) is

  1. 25mg Bromophenol Blue
  2. 3 ml of glycerol
  3. Add water to 10 ml

Loading Buffer with GelRed for Electrophoresis


To do this buffer you can use either Bromophenol blue or Xylene Cyanol buffer described above. This protocol will make 5ml of 6x solution. Adjust to your needs if necessary.

  1. 5 ml Loading Buffer (Xylene-cyanol or Bromophenol Blue)
  2. 15 ul of GelRed (10000x in water, cat.41003 Biotium)


1 Kb Ladder for Electrophoresis


Use the ThermoFisher GeneRuler 1kb ladder or GeneRuler 1kb plus from the stock. Mix:

  1. 100µl (1 tube) DNA Ladder
  2. 100µl 6X Loading Dye Solution
  3. 400µl Deionized water

Good to go. Load 3 to 5 µl.
