Kdp screening Many transformant colonies were found on chloramphenicol plates. Single colony PCR was perfomed over a selection of 44 colonies using forward and reverse biobrick primers in order to gauge the size of the vector insert by gel visualisation. All screened colonies were negative for Kdp - concluded most likely to be blunt ended self ligation of pSB4C5 vector (without death gene).
