8/18 TesA+RBS: Transformed ligation/- control that was in freezer. +Control: mCherry 3 μL 200 μL of mixture on chlor and 200 on amp. ACC Plasmid: Ran a gel of amplified plasmid parts to ensure that the PCR worked. -Gel looked good, but was very faint. bands were in the proper places. Ethanol: Re-transformed PACYC plasmid into BL21 cell line.
