LCR PCR Colony Screen

Purpose: To screen for proper insertion of Gal4DB_mCh and ATF2 into the MV10 vector.

Methods:

GoTaq Colony PCR (1RXN):

  • 5uL 2XGoTaq
  • 0.5uL FP (CMV fwd)
  • 0.5uL RP (DD123)
  • 4uL Water

Controls: - control of water only, MV10

6 colonies selected from LCR1 and 6 colonies from LCR2 Incubated in 10uL of mastermix for 10min at RT Run on GoTaq PCR program (60C, 30min)
