Safety measures
- When working with yeast - work only in the biological hoog - for your safety!
Protocol
- measure 600 ml DDW in a water bottle
- Optional add HCl until reaching pH 3.5-4
- Put the bottle on the stirrer with heating and add slowly 30 gr of YPD broth
- Put the bottle in autoclave bag
- Autoclave the bottle, 'make sure not the close the cup completely
- after cooling down, add 100 ul of yeast into the bottle
- put in incubator with shaker on 28-30oc for 48 hours
