Steve Koch 01:36, 27 January 2010 (EST): I just looked at the gel photo in the dark and from an angle and it didn't look too bad. Maybe it's a full moon? I think you should post the original photo + an inverted, contrast-adjusted photo from now on. That would make it easier to see.
I definitely think the reaction worked, but not sure why it looks like it's >100 bp? I worry about the high voltage still, but don't know how that could matter, given that the 100 bp ladder looks OK. But, definitely that looks like a product band. Can you run it through a QIAquick to get rid of primers and then run a gel?
