Targetron msa knockout
- Plasmid prep from msa/control , RN4220 and Dh5
- Gel shift
- 4220 didnt work out
- Dh5 worked out, all colonies picked had the plasmid
Transformation
- Used the Dh5 derived retargeted plasmed to transform 4220
- Plated both msa/control on erm, incubated at 32oC
Cell wall
- Isolate cell wall from wt/mutant
- Cells grown overnight in TSB, without antibiotics at 225 rpm and 37oC
