The objective of this experiment is to establish the biomineralization of gold nanoparticles in Bovine Serum Albumin such that the gold nanoparticles can be used effectively in future experiments.


Protocol: Between 0.0015 and 0.015 mM of aqueous Bovine Serum Albumin (BSA) was mixed with between 0.25 and 1.0 mM of aqueous Chloroauric Acid (HAuCl4) at room temperature. Once the mixture was complete, separate samples of the mixture were kept at 40, 60, and 80 degrees Celsius in hot water baths for 6 hours. pH measurements were taken periodically over the course of the 6 hours for each sample at each temperature. The samples were also monitored between 20 and 70 degrees Celsius to determine how the denaturization of BSA affects the creation of the gold nanoparticles. The samples changed from colorless to pink and finally to purple over the course of the 6 hours in the hot water baths. UV-visible spectra analyses were taken of each sample with a UV Spectrophotometer in the wavelength range of 200 to 900 nm to determine the absorbance of each sample. The samples were then cooled to room temperature and kept overnight. The next day, the gold nanoparticles were collected by washing the samples with distilled water and then centrifuging them at 10,000 to 14,000 rpm for 5 minutes. This nanoparticle collection was done twice or more to ensure all possible nanoparticles were extracted. BSA 1.5mM-->0.0013grams Au3+ 0.25mM-->0.0007grams fill to 10mL with Tris 7.55pH


I would also like to see this graph zoomed in to the features at 550. The thing I would like to know is: Are the changes at 550nm due to a new feature in the spectrum or just an increase in the baseline<b> Matt Hartings 08:50, 6 September 2011 (EDT)
