Objective

Running Mb catalyzed H2O2 and o-phenylenediamine reaction today.

Procedure and Data

Stock solutions

  1. Myoglobin
    1. 2.0mg (Mb:KCl lyophilized from Citrate, 5mg:500mg) in 1.0mL water
  2. OPD
    1. 11.5mg OPD in 11.5mL water
  3. H2O2
    1. 0.227mL 30% H2O2 in 1.773mL water

Sample 1

  1. Sample chamber set to 25C and to stir
  2. 2.025mL OPD
  3. 0.75mL H2O2
  4. Start Data Collection
    1. 1ms Integration
    2. 10 scan average
    3. Wait 1 minute after each scan
    4. Stop after 2 hours
    5. Files saved as: Mb_OPD_H2O2_Citrate_H20_25_SCANNUMBER.txt
  5. 0.225mL Mb added just before the third scan
    1. This is denoted in the data file for the third scan

Sample 2

  1. Sample chamber set to 15C and to stir
  2. 2.025mL OPD *Note---> The OPD had already started to turn yellow in the container. Must make a fresh sample for each run. This is probably what went wrong with Dhea and Paul's stuff this summer. Matt Hartings
  3. 0.75mL H2O2
  4. Start Data Collection
    1. 1ms Integration
    2. 10 scan average
    3. Wait 1 minute after each scan
    4. Stop after 2 hours
    5. Files saved as: Mb_OPD_H2O2_Citrate_H20_15_SCANNUMBER.txt
  5. 0.225mL Mb added just before the third scan
