Objective Prepare media for hemoglobin expression and set up starter cultures. Procedure Starter cultures (x4) 0.875g LB 35mL water autoclave Expression cultures (x4) 25g LB 1L water autoclave Starter cultures (x4) 35uL of 100mg/mL ampicilin was added to each starter culture flask Starter culture was inoculated with cells from the BL21(DE3)/pET-3d/WT Ascaris hemoglobin glycerol stock Flasks were placed in the shaker/incubator overnight (37C, 250rpm)
