Objective

Using previous experiments as a guide, another AuNP synthesis was run with the goal of determining what the AuNP fibers are stable in and to prevent BSA protein unfolding.

Procedure

Two solutions of AuNP were created. Solution 1:

  • 0.516 mL of 30 μM BSA was combined with 0.350 mL of 4.17 mM HAuCl4 in a test tube.

Solution 2:

  • 0.516 mL of 30 μM BSA was combined with 0.700 mL of 4.17 mM HAuCl4 in a test tube.

When Solution 1 test tube began to turn purple, it was removed from the oven and 100 uL of 5.02 mM tris (pH 10.1) was added to 1 mL of the solution. The solution then underwent UV VIS analysis. 100 uL of 5.02 mM tris was added two more times, and after both times UV VIS was also used to analysis the solution.

Results

Data for Solution 1 (UV-VIS):

Purple fibers were observed in Solution 2.
