Objective The purpose of today's experiments were to make a labeled cysteine fluorophore and to measure the conductivity of the purple fibers previously created. Protocol Fluorophore: 2 mg of 5-iodoacetamido fluorescein (5-IAF) were combined with excess cysteine (0.39 mg) in 3.95 mL of 50 mM Tris buffer (pH 7.5). The solution was vortexed to mix. The reaction tube was wrapped in tin foil an d left for 2 hours. 100 μL of dithiothreitol (DTT) was added to the reaction tube. Results Conductivity: The conductivity testing was unsuccessful because the Pd wire did not adhere to the Teflon plate.
