Objective

The purpose of today's experiments were to make a labeled cysteine fluorophore and to measure the conductivity of the purple fibers previously created.

Protocol

Fluorophore:

  • 2 mg of 5-iodoacetamido fluorescein (5-IAF) were combined with excess cysteine (0.39 mg) in 3.95 mL of 50 mM Tris buffer (pH 7.5).
  • The solution was vortexed to mix.
  • The reaction tube was wrapped in tin foil an d left for 2 hours.
  • 100 μL of dithiothreitol (DTT) was added to the reaction tube.

Results

Conductivity:

The conductivity testing was unsuccessful because the Pd wire did not adhere to the Teflon plate.
