October 14th, 2010

1. Suspend in liquid LB medium 8 colonies of transformation:

  • pSB4A5 + MinBP + ΔRBS + GFP E004

These colonies will be used for plasmid extraction.


2. Extract plasmid from 8 colonies suspended in the last step, plasmid extraction performed with the High Pure Plasmid Isolation Kit Roche.


3. Resuspend in liquid LB medium with correspondent antibiotic:

  • pSB4A5 + MinBP + ΔRBS + GFP E004
  • I20269 (pSB3K3 + J23101 + GFP E0040)

This cells will be tested under microscope for fluorescence.
