Ligation:
vol insert = vol backbone * (C_backbone / C_insert) * (n_insert / n_backbone) * (bp_backbone / bp_plasmid) / (BP_backbone / BP_insert)
The above formula is valid when the backbone-bearing plasid is digested, treated with CIP and used in the ligation (no isolation of backbone fragment). BP_plasmid is the length of the complete plasmid, and BP_backbone is the length of the fragment to which the insert is ligated.
