Concerning news Ok, so nothing really worked out:( I came in early and check all my plates. Only one plate showed any suspicion of growth - the standard NEB ligated. I am surprised but again not - as I was expecting this to happen. At first I thought it might have been my Kanamycin as my viability test failed. The viability is basically show whether or not the standard plasmid would transform - in other words it is just a transformation test. I am still not sure why noting grew. I thought it might have been the Kanamycin as I was making a stock at 50mg/ml whilst the other group was using only 10mg/ml. I checked out a couple of website - and it seems fine - I will adjust to my other groups to see if growth improves. Concerning news regarding my future establishment in the lab....I hope it will work out!
