Objective

  • Perform a redox titration on HRP in order to determine the standard potential of this protein.

Procedure

Each group will do 6 measurements. 2 groups will start off with 30uM HRP solutions (with some redox mediators). 3 groups will start off with 30uM HRP solutions (with some redox mediators) and some added ferricyanide. All groups will add 1uL or 2uL of concentrated sodium dithionite and observe both the UV-Vis spectrum as well as the galvanic cell potential. This measurement will be repeated by each group 6 times.
