(ng/µL) St Dev (ng/µL) 260/280 SETD7 1** 27.00 3.20 1.99 SETD7 2** 24.418 3.63 1.98 ANM5 1* 216.875 5.04 1.90 ANM5 2* 312.5 4.13 1.89 Conclusion This is the results from the third miniprep for SETD7 plasmid that was purchased from DNASU, this is also the second run for the ANM5 plasmid. Prior results showed the similar 27ng/µL concentration of SETD7 and I wanted to run more tests to see if I could increase the yield, however this has not been the case. I have decided to use the transformed cells and re-plate them to see if it was just the first plating that was the issue. This is the second run for the ANM5 plasmid and I was able to get a greater concentration of plasmid using 10mL of LB amp broth than the 5mL. I still need to run a restriction digest to prove that the insert is in the plasmid.
